Ganoderma lucidum is the dried fruiting body of Polyporaceae fungus Ganoderma lucidum or purple lucidum. It has the functions of invigorating qi and soothing the nerves, relieving cough and asthma, and anti-tumor. More than 300 active compounds have been isolated from Ganoderma lucidum, including triterpenoids, polysaccharides, alkaloids, fatty acids, steroids, lactones, peptides, oligosaccharides, and trace elements. Triterpenoids and polysaccharides are considered to be the main components of Ganoderma lucidum to exert anti-tumor effect.
The researchers explored the anti-lung cancer effect and mechanism of Ganoderma lucidum ethanol extract. They used CCK-8 method to determine the growth inhibition rate of Ganoderma lucidum ethanol extract on the growth of lung cancer A549 cells; observed the effect of Ganoderma lucidum ethanol extract on cell morphology by fluorescence microscope; observed its effect on cell cycle by flow cytometry; used AnnexinV- FITC/PI double staining was used to detect the apoptosis of cells.
When the concentration of Ganoderma lucidum ethanol extract was 5-320 μg·mL-1, the growth inhibition rate of A549 cells gradually increased (see Table 1), and the half-inhibitory concentration was 10.8 μg·mL-1. Under the fluorescence microscope, the A549 cells treated with different concentrations of Ganoderma lucidum ethanolic extract showed cell shrinkage, chromatin edge agglutination and other specific morphological characteristics of apoptotic cells.
When the concentration of Ganoderma lucidum ethanol extract was 80, 100, and 200 μg·mL-1, the cell cycle was mainly arrested in G0/G1 phase. When the concentration of Ganoderma lucidum ethanol extract was 300, 350 μg·mL-1, the cell cycle was mainly arrested Periodic arrest is in S phase (see Table 2). When the concentration of Ganoderma lucidum ethanol extract was 200, 300, and 350 μg·mL-1, the early apoptosis rates were 21.66%±5.19%, 27.34%±2.89%, 28.28%±7.25%, respectively, and the apoptosis rates increased significantly, and Upward trend.
Experiments show that Ganoderma lucidum ethanol extract has a good anti-lung cancer effect. The main anti-tumor effect on lung cancer cells may be achieved by inhibiting the proliferation of lung cancer cells A549, affecting cell morphology, blocking cell cycle and inducing cell apoptosis.
